Luke and his twin sister, Leia, are born to Padmé Amidala on Polis Massa. Their father is Anakin (Darth Vader) Skywalker.

Star Wars Episode III: Revenge of the Sith
Padmé dies after giving birth to Luke and Leia. Jedi Masters Obi-Wan Kenobi and Yoda decide that they must hide the Skywalker children from their father, former Jedi Knight Anakin Skywalker, who had recently turned to the dark side and became the Sith Lord Darth Vader. Leia is sent to Alderaan with the droids R2-D2 and C-3PO with her adoptive parents, Bail and Breha Organa, so she would not be found by the Empire. Luke would be taken to the desert planet of Tatooine to be raised as a moisture farmer by his Uncle Owen and Aunt Beru. Obi-Wan would then go into exile on Tatooine to keep an eye on Luke during his formative years.

Star Wars Episode IV: A New Hope
Luke and Leia meet for the first time when Luke (along with Obi-Wan and Han Solo) board the Death Star where Leia is being held captive by Darth Vader. Earlier in the film, Luke had bought the two droids R2-D2 and C-3PO and found the help message addressed to Obi-Wan from Princess Leia, speaking of her capture and need for rescue. Luke eventually leaves with Obi-Wan and Han Solo for the Death Star to rescue Princess Leia. Luke successfully rescues Leia and then joins the Rebel Alliance and destroys the Death Star. Neither Luke nor Leia know at the time that they are twin brother and sister.

Star Wars Episode V: The Empire Strikes Back
Luke and Leia are still unaware of their relationship of brother and siser and share a kiss, however romance blossoms between Leia and Han Solo. Leia, while with Han, stops at Bespin for repairs. While there Han's friend, Lando Calrissian, turns them over to Darth Vader, who uses them as bait for Luke. Luke is lured into a lightsaber duel with Darth Vader. As his mentors warned, Luke proves to be no match for Vader, who cuts off his right hand. The Sith Lord then reveals his true identity as Luke's father. Vader offers him the chance to turn to the dark side of the Force and rule the galaxy at his father's side. Horrified by this unexpected claim and the powerful temptation before him, Luke chooses to die rather than be corrupted and throws himself into a deep reactor chasm. However, he is sucked into a garbage chute to the underside of Cloud City and left hanging onto a weather vane. Leia, flying away from Cloud City in the Millennium Falcon, senses Luke's peril and turns the ship around to save him.

Star Wars Episode VI: Return of the Jedi
Leia in an attempt to free Han Solo from Jabba the Hut is caught and turned into Jabba's slave girl. Luke (now nearly a Jedi Knight) offers to negotiate with Jabba, but instead is forced to fight a fearsome monster, the Rancor. When Luke kills it, Jabba sentences him to be thrown into The Pit of Carkoon, to be eaten by the Sarlacc. Luke escapes with R2's help, saving his friends and destroying Jabba's barge, with the aid of a new green lightsaber constructed following the loss of his first weapon in his duel with Vader. During Luke's return trip to Dagobah, Luke learns from the dying Yoda that Vader is indeed his father, and from Obi-Wan's spirit that he has a twin sister, which Luke immediately realizes is Leia. While preparing for a last battle with the Empire on Endor, Luke reveals to a stunned Leia that she is, in fact, his twin sister, and that Vader is their father. Initially reluctant to believe him, Leia realizes that Luke speaks the truth and joins Han Solo in leading the Rebels in battle on Endor as the Rebel Fleet battle the second Death Star. Later Luke is trying to be turned to the dark side by Darth Vader and the Emperor. While in a battle with Darth Vader, Luke is keeping his emotions under control until Vader senses Luke's feelings for his sister, Leia, and threatens to turn her instead. At this threat Luke finally gives into his anger and attacks with full fury. Vader loses ground as Luke lands blow after blow; Luke knocks Vader to the ground and severs his father's mechanical right hand. With the Emperor ordering Luke to strike Vader down and take his place, Luke glances at his own bionic hand and realizes with a visceral horror that he's on the verge of suffering his father's fate. Luke regains composure, deactivates his lightsaber and throws it away, proudly declaring his allegiance to the Jedi. Enraged, The Emperor attacks Luke with Force lightning, nearly killing him. In horrible pain, Luke calls out to Vader for help. In an act of self-sacrifice, his father seizes the Emperor and throws him to his death down a reactor shaft. In the process, he is mortally wounded by the Emperor's lightning. As Rebel fighters and the Falcon race toward the Death Star's main reactor, Luke (at his father's request) removes the mask that had encased his face, and looks upon his father's eyes for the only time. A redeemed Anakin Skywalker dies moments later, assuring his son that there is still good in him after all. Luke escapes with his father's remains moments before the Death Star is obliterated. Luke then returns to Endor, where he is reunited with Leia.
